How to use the platform

LumiPoreTM nanoparticle transfection workflow

Suitable for R&D and clinical applications
 
  1. Add nanosensitizers to cell culture medium and leave to incubate (1-30 min)
  2. Add payload of interest to cell culture medium
  3. Perform laser irradiation on LumiPoreTM platform (<10 min. for full 96-well plate)
  4. Optional: wash and add fresh cell medium

Suitable for cell therapy manufacturing and high-throughput applications
 
  1. Either culture cells or add cells in suspension to plate with embedded nanofibers and let cells sediment (1-10 min)
  2. Add payload of interest to cell culture medium
  3. Perform laser irradiation in LumiPoreTM platform (<10 min. for full 96-well plate)
  4. Optional: wash and add fresh cell medium

Advantages of using the LumiPoreTM Platform

Excellent cell viability

The LumiPoreTM technology is very gentle to cells, ensuring minimal impact on cell viability, cell proliferation, homeostasis and functionality. This has for instance been demonstrated on hard-to-transfect human T cells, which proliferate almost immediately and maintain their full cell-killing potential after transfection with the LumiPoreTM system.

Proliferation of activated T cells after transfection by photoporation happens equally as fast as non-treated T cells, while electroporated T cells show delayed proliferation. Reference: Xiong et al. Nat Nanotechnol (2021)

Improved yield

The LumiPoreTM platform is a gentler technology than the current state-of-the-art electroporation technologies and therefore improves the transfection yield, resulting in a higher number of transfected and viable cells. For instance, photoporation leads to 2.5 times more living mRNA-transfected stimulated human T cells as compared to the current gold standard electroporation method. Reference: Harizaj et al. High throughput

The LumiPoreTM technology offers high throughput with limited hands-on time, making it suitable for a large variety of applications including screening assays. After adding the proprietary nanosensitizers and effector molecules to the cells, the recipient is placed in the device and the transfection can be started with a simple click of a button. An entire multi-well plate is transfected in less than ten minutes.

Advanced cell targeting

LumiPoreTM offers the unique possibility to transfect specific subpopulations of cells. For instance, cells can be transfected according to user-defined spatial patterns. This opens up unique experimental solutions for tissue engineering applications or studying bystander effects.

Additionally, the LumiPoreTM platform can optionally integrate transmission and fluorescence imaging of cells. This allows for the delivery of payloads to specific cells which are either selected manually or automatically by making use of automated in-line image processing. Transfection can be done with single cell resolution.

Illustrated here is the selective delivery of two different fluorescent labels to adjacent neurons, facilitating interaction studies between these cells.

Flexibility

The LumiPoreTM platform offers efficient cytosolic delivery of various payloads, including nucleic acids, proteins and contrast agents. It works efficiently on a broad range of cell types, including hard-to-transfect cells such as stem and immune cells.
