pDNA transfection
Efficient delivery of exogenous DNA into mammalian cells remains a critical challenge in cell engineering, requiring a careful balance between high transfection efficiency and preservation of cell viability, phenotype, and functionality.
Since mammalian cells do not naturally take up plasmid DNA (pDNA), specialized intracellular delivery technologies are required to introduce genetic material into the cell. As a result, transfection efficiency often becomes a key bottleneck, directly impacting protein expression levels, cell engineering outcomes, and overall workflow performance.
In addition, workflows involving stable cell line generation require further selection and single-cell cloning steps following transfection to isolate consistently high-expressing clones, increasing both complexity and development timelines.
Gentle Photoporation with LumiPore®
The LumiPore platform addresses these challenges through a gentle photoporation mechanism that enables efficient pDNA delivery while minimizing cellular perturbation. Unlike conventional physical transfection methods, LumiPore preserves cell viability and functionality, allowing cells to recover rapidly after treatment.
This combination of efficiency, gentleness, and scalability makes LumiPore particularly well suited for protein production, stable cell line generation, and other pDNA-based applications where both cell health and productivity are critical.
Application Datasets & VIB Collaboration
The following sections present representative datasets showcasing transient and stable pDNA transfection in Expi293 and ExpiCHO cells using the LumiPore platform. Generated in collaboration with the VIB Protein Core (Ghent, Belgium), these results demonstrate the potential of gentle and scalable photoporation for applications ranging from rapid protein expression to stable cell line generation.
Further details on experimental conditions, validated protocols, and best practices are provided in the corresponding sections below.
